Standard Operating Procedure for glassware cleaning and its sterilization
1.0 PURPOSE:
To lay down a procedure for glassware cleaning and its sterilization.
2.0 SCOPE:
This procedure is applicable for glassware cleaning and its sterilization in Microbiology Laboratory.
3.0 RESPONSIBILITIES:
Microbiologist: To perform the procedure of glassware cleaning and its sterilization as per SOP.
Head-QC: To review and implementation and ensure compliance of SOP.
Head-QA: To approve, implement and ensure compliance of SOP.
4.0 PROCEDURE:
4.1 Glassware’s like Measuring cylinder, petri plate, conical flask, glass-rod, beaker, test-tube etc. used for the general purposes like Media preparation, titration, reagent making etc., in which there have not become dirty, so they are immediately rinsed after use with the tap water several times and then can be washed according to the method described below.
4.2 Those glassware which have been used for bacteriological analysis works or which have become dirty due to the used media, live culture or other substance needs special procedure for wash.
4.3 If such glassware contains infected material they should first be autoclave and their content must be removed properly.
4.4 All such glassware is then soaked in chromic acid solution for overnight and later on they are rinse with tap water and then with Purified water.
4.5 Procedure for Glassware Washing:
4.5.1 After acid treatment and autoclaving of dirty glasswares, transfer all the glasswares in another tub containing mild soap solution (1%-2% solution).
4.5.2 Soak all the glasswares for one hour & scrub each of the glassware thoroughly with brush.
4.5.3 Wash all glassware under running tap water several times to remove all traces of detergent.
4.5.4 Finally rinse with purified water for at least three times.
4.5.5 After that keep all the glasswares into the oven at 60°C.
4.5.6 For sterilization of glasswares, wrap them with aluminum foil.
4.5.7 Glassware is kept 1 hour in oven at set temperature 180°C for dry heat sterilization or run autoclave cycle at 121°C for 30 minutes for moist heat sterilization.
