Standard Operating Procedure for efficacy check of membrane filter used in filtration method in testing of purified water

1.0   PURPOSE:

To lay down a procedure for efficacy check of membrane filter used in filtration method in testing of purified water.

2.0   SCOPE:

This procedure is applicable for efficacy check of membrane filter in Microbiology Laboratory.

3.0   RESPONSIBILITIES:

Microbiologist: To perform Efficacy check of membrane filter as per SOP.

Head-QC: To review and implementation and ensure compliance of SOP.

Head-QA: To approve, implement and ensure compliance of SOP.

4.0   PROCEDURE:

4.1    Every lot of membrane filter should be check for its Efficacy test for surety of filter paper.

4.2    Take four filter paper and four sterile filtration assemblies.

4.3    Place all membrane filter papers which are under test on a membrane filter holder. Wet the membrane filter with 100ml of sterile Buffered sodium chloride peptone solution (pH 7.0) or 100ml peptone water.

4.4    Now transfer 1ml of pure culture (10-100 CFU/ml) of Bacillus subtilis (ATCC 6633), Candida albicans (ATCC 10231), Escherichia coli (ATCC 8739), and Pseudomonas aeruginosa (ATCC 9027) individually on separate filter paper.

4.5    Rinse with 100ml of sterile Buffered sodium chloride peptone solution (pH 7.0) or 100ml peptone water.

4.6    Collect filtrate and add 10ml filtrate to 100ml sterile Soyabean Casein Digest Medium. Incubate it at 30-35°C for 14 days.

4.7    Rinse filter paper again with 100ml of sterile Buffered sodium chloride peptone solution (pH 7.0) or 100ml peptone water twice.

4.8    Take filter paper and incubate all in separate 100ml sterile Soyabean Casein Digest Medium and incubate it at 30-35°C for 7 days.

4.9    Perform the blank test prior to the above test in which instead of mix culture suspension, filter 10 ml of sterile Rinse solution (Peptone water 0.1% or BSCP) through the membrane filter.

5.0    Incubate filtrate and filter paper same as above.

5.1    Note:No turbidity shall be observed till 14 days in tubes containing the filtrate. Growth shall be observed within 7 days in the tubes containing membrane filter  and Blank or negative control tubes should not show any turbidity.
