Standard Operating Procedure for Disposal of growth containing media and culture
1.0 PURPOSE:
To lay down a procedure for Disposal of growth containing media and culture.
2.0 SCOPE:
This procedure is applicable for Disposal of growth containing media and culture in Microbiological Laboratory.
3.0 RESPONSIBILITIES:
Microbiologist : To perform the procedure of disposal of growth containing media and culture as per SOP.
Head-QC : To review and implementation and ensure compliance of SOP.
Head-QA : To approve, implement and ensure compliance of SOP.
4.0 PROCEDURE:
4.1 The laboratory waste includes discarded culture, used media, used cotton plugs etc; all such wastes are the potential source of pathogen.
4.2 Specimens, cultures and other materials which may contain infectious pathogens and equipments used to handle such materials must be made non infectious before being discarded or cleaned for reuse.
4.3 Moist steam under high pressure is the most effective means of the achieving sterilization.
4.4 Prepare 2% of Savlon, Dettol or any other disinfectant solution on weekly rotation basis in the large beaker or bucket.
4.5 Transfer the waste media or culture from glass ware to bucket and immersed in prepared disinfectant solution as per rotation basis. Immersed for 30 min.
4.6 All media treated with 2% disinfectant transfer into autoclavable disposable bag. Wrap tightly with rubber band or staple.
4.7 Place autoclavable Bag containing waste culture or used media in the large bucket.
4.8 Operate the Autoclave on 15psi, 121oC for 30 minute as per SOP for Operation and cleaning of vertical autoclave for destruction.
4.9 Put a chemical strips or indicator for sterilization to check effectiveness of sterilization.
4.10 Send this discarded material to the ETP (Effluent Treatment Plant).
4.11 Clean the Glass ware as per SOP for glass ware cleaning and its sterilization.
4.12 Washing shall be done very thoroughly with purified water so that media residue does not remain in the Glass wares.
