SOP for Personnel Monitoring by contact plate method in controlled area.
1.0 Purpose:
To lay down a standard operating procedure for personnel monitoring by contact plate method.
2.0 Scope:
This procedure is applicable for personnel monitoring by contact plate method in Microbiology Laboratory.
3.0 Responsibility:
Microbiologist / Designee:
• To follow personnel monitoring by contact plate method as per SOP.
• To follow personnel monitoring by contact plate method as per SOP.
• To perform the sampling and monitoring of contact plate as per frequency.
• To observed the plate as per incubation period.

4.0 Procedure:
4.1 General Procedure:
4.1.1 Prepare soyabean casein digest agar media and sterilize it as per SOP for media preparation.
4.1.2 Prepare RODAC plates (Diameter 55 mm) by pouring sufficient soyabean casein digest agar. Pour the plates in such a way that the surface of the medium is slightly raised in comparison to the edge of the plate.
4.1.3 Allow the medium to solidify.
4.1.4 Incubate the plates at 30-35°C for about 48 hours before use.
(Preincubation)
4.1.5 Observe the plates for convex surface and for any growth. If any plate is found contaminated discard it as per SOP for Disposal of growth containing media and culture.
4.1.6 Aseptically transfer the plates to respective area, where the sampling has to be carried out.
4.2 Sampling Procedure for Personnel Monitoring:
4.2.1 Perform the personnel monitoring in controlled area from the following gown locations:
• Head gear
• Middle of the Chest
• Right forearm
• Left forearm
• Palm
• Head gear
• Middle of the Chest
• Right forearm
• Left forearm
• Palm
4.2.2 Open the lid of plate and gently contact (touch) the plate over the gowning location to be monitored.
4.2.3 The plate should be first incubated at 20-25°C for 3 days (fungi) and 30-35°C for 2 days (Bacteria).
4.2.4 Count to be recorded at the end of 5th day. Simultaneously also place one positive and negative control with test.
4.2.5 Count the number of colonies each plate, and record the number of colonies as cfu /gown location.
4.2.6 Acceptance Criteria:
