Standard operating procedure for Operation and Calibration of antibiotic Zone Reader
1.0 Purpose:
To lay down a procedure for Operation and calibration of zone reader.
2.0 Scope:
This procedure is applicable for operation and calibration of Zone Reader having Make- DBK Instrument in Microbiology laboratory.
3.0 Responsibility:
Microbiologist/ Designee:To operate, calibration and maintain the record as per SOP.
Head/ Designee - QC :To review and ensure compliance of SOP.
Head/ Designee - QA :To approve, implement and ensure compliance of SOP.
4.0 Procedure:
4.1 Precaution:
4.1.1 All the electric wires should be properly insulated.
4.1.2 Only authorized persons should handle the instrument.
4.2 Operation:
4.2.1 Connect the three pin plug to the main supply.
4.2.2 Switch "ON" the main switch. Digital display will be showing some reading.
4.2.3 Switch "ON" the Lamp.
4.2.4 Turn the right side 'Slider knob' fully anticlockwise.
4.2.5 Keep the sample Petri dish on the top belt.
4.2.6 Put magnetic clamp around the petri dish and slide it manually.
4.2.7 One edge of circle zone image just touches the mark line on the viewing glass.
4.2.8 Rotate the dial given below the display unit to focus the image of the sample.
4.2.9 If required, again adjust the arc precisely from one edge of zone, tangential with mark line on the viewing glass by the 'slider knob'.
4.2.10 Press "ZERO" switch for few seconds and then release to make the reading 00.0 on the display.
4.2.11 Now turn the right side slider knob slowly, in clock wise position, till the other edge of the zone image touches the mark line on the viewing glass.
4.2.12 Note the reading in millimeter (mm) directly on the digital display.
4.3 Calibration:
4.3.1 Take the calibrated circular disc.
4.3.2 Place the disc between the magnetic holders.
4.3.3 Adjust scale to zero and move the scale so that the entire disc is covered.
4.3.4 Record the data in calibration record format.
4.3.5 Measure the diameter of the disc by Vernier caliper and record the reading in calibration record format.
4.3.6 Acceptance criteria: ±2 mm of measured diameter.
4.3.7 Frequency: Once in a 3 month.
4.4 Maintenance:
4.4.1 If the instrument does not produce required operation results, its response is poor then it should be labeled as "UNDER MAINTENANCE" and should be repaired or serviced.
