Standard Operating Procedure for Cleaning and Disinfection of Microbiology area

1.0 Purpose:
To lay down a procedure for cleaning and disinfection in Microbiology Laboratory.
2.0 Scope:
This procedure is applicable for the cleaning and disinfection of Microbiology Laboratory.
SOP for Cleaning and Disinfection of Microbiology area

3.0 Responsibilities:
Housekeeping Supervisor/Laboratory Assistant:
•To clean the microbiology area and equipment as per procedure.
Microbiologist/Designee:
•To ensure cleaning and disinfection procedure as per SOP.
•To prepare the disinfectant as per schedule.
•To maintain the record as per SOP.
QC Head/Designee :
•To review implementation and ensure compliance of SOP.
QA Head/Designee:
•To approve, implementation and ensure compliance of SOP.
4.0 Procedure:
4.1 Precaution:
4.1.1 Use proper safety apparatus such as gown, hand gloves and safety goggle during preparation and use of disinfectants.
4.1.2 All disinfectant solution shall be prepared freshly.
4.1.3 Use clean duster for cleaning of microbiology lab and its Bioassay room-I, bioassay room-II, change rooms, Airlocks, media preparation room, autoclave room and incubator room.
4.1.4 Always add disinfectant to purified water while diluting.
4.2 Disinfectant Solutions:
4.2.1 Prepare quantities of disinfectant solution sufficient for one day cleaning (Dettol 2.5%, Savlon 2.5% and Triple 256; 2.5%).
4.2.2 70% IPA for hand disinfection and cleaning for 3 days.
4.3 Disinfectant Solution Preparation:
4.3.1 Dettol 2.5%:
4.3.1.1 For examples 1000 ml: Take 500 ml of purified water add 25 ml of dettol and make up volume to 1000 ml with purified water. Mix well and use within a day.
4.3.2 Savlon 2.5%:
4.3.2.1 For examples 1000 ml: Take 500 ml of purified water add 25 ml of savlon and make up volume to 1000 ml with purified water. Mix well and use within a day.
4.3.3 Triple-256 ; 2.5%:
4.3.3.1 For examples 1000 ml: Take 500 ml of purified water add 25 ml of triple -256 and make up volume to 1000 ml with purified water. Mix well and use within a day.
4.3.4 70% Isopropyl Alcohol:
4.3.4.1 For examples 1000 ml: Take 700 ml of IPA add 300 ml of purified water. Mix well and use within 3 days.
4.3.5 Preparation of all disinfectant solutions (other than IPA) shall maintain as per record.
4.3.6 70 % IPA preparation & distribution record shall maintain as per record.
4.3.7 Disinfectant label shall be affixed as per format.
4.4 Cleaning Procedure of Floor:
4.4.1 Floor of the lab area must be cleaned with savlon 2.5%, dettol 2.5% and triple-256; 2.5% daily in the morning.
4.4.2 Daily mop the floor with help of mopper.
4.4.3 Daily in the morning or after the work is over; clean the entire working area as per format.
4.4.4 Daily mop the floor surface with disinfectant given below. The sequence of floor cleaning is as per the rotation of disinfectant.
4.5 Cleaning Procedure of Door, Glass Panels and Wall:
4.5.1 Mop the wall surfaces with disinfectant solutions as mentioned below.
4.5.2 For cleaning of door, glass penal 70% IPA solution should be used.
4.5.3 Cleaning done early in the morning.
4.5.4 Frequency: Cleaning of walls and doors is once in a week.
4.6 Cleaning Procedure of Ceiling/Grills:
4.6.1 Climb on a ladder to clean the ceiling and grills.
4.6.2 Use safety goggles to prevent dust falling into the eyes.
4.6.3 Cleaning done early in the morning.
4.6.4 Wipe out the entire ceiling with duster.
4.6.5 Frequency:  Cleaning of ceiling and grills is once in a month.
4.7 Disinfection of Hands:
4.7.1 Use 70% IPA for disinfecting the hands.
4.8 LAF and Pass Box:
4.8.1 Daily cleaning LAF bench and pass box with 70% IPA solution.
4.9 Instruments:
4.9.1 Daily wipe the outer surface of all instruments with 70% IPA solution with a lint free cloth.
4.9.2 Weekly cleaning the inner surface of all equipment or instruments with dettol 2.5%, savlon 2.5% and triple-256; 2.5% solution.
4.9.3 Instrument cleaning done every day by outside and weekly from inside with proper disinfectant as per format.
4.10 Waste Bin:
4.10.1 Collect waste material in polythene bags from the waste bins and tie it properly with a nylon thread and send it to the scrap yard.
4.10.2 Daily clean the inner and outer surface of the waste bins with a dry lint free cloth.
4.10.3 Weekly wash the waste bins with savlon 2.5%, dettol 2.5% and triple-256; 2.5% solution.
4.11 Disinfection of Mop:
4.11.1 Daily before mopping, clean the mop with fresh water.
4.11.2 After the completion of mopping activity clean the mop and dipped in dettol 2.5%, savlon 2.5% and triple-256; 2.5% solution for 15 min.
